I've never known or understood what the difference in these two sliders was. I took a Photoshop class a while back and even asked my instructor (who was a great instructor) and he didn't know either!
The issue has always been something I have wondered about but hadn't really ever researched. Well I finally learned what the difference is between these two options. You work with them and they appear to do almost the same thing. And it's true they are very similar but the difference is this. Opacity will lower the opacity of the entire layer. Fill will lower the opacity of the pixels on the layer as well, but not the layer style effects you have added to the layer. Try it out! Move an element or something onto your layout (then give it a drop shadow) and lower the "fill" slider. If you move it all the way to the left - to 0 the shadow will still be there, but the element will not show.
